If you place Sys.WebForms.PageRequestManager.getInstance() in certain areas of ASPX page you may get an error.

Especially when you are using ScriptManager or UpdatePanel, you will have to call ASP.NET AJAX before loading PageRequestManager.

The following code shows where I placed the script. As for now, it is functioning fine.

<asp:ScriptManager ID="scriptManager" runat="server" >
</asp:ScriptManager>

            <script type="text/javascript">
            var Instance = Sys.WebForms.PageRequestManager.getInstance();
          </script >

<asp:UpdatePanel ID="UpdatePanel1" runat="server">
